Lincosamides

Lincosamides - The lincosamides include lincomycin and clindamycin, of which the former has been more extensively studied. Lincomycin is active against many gram positive bacteria, but is inactive against Bram negative species. Its spectrum, therefore, resembles that of penicillin. It does not inhibit protein synthesis in mammalian cells, and shows no activity against 80S ribosomes. It binds exclusively to the 50S subunit of 70S ribosomes and inhibits.

peptide bond synthesis. Lincomycin causes breakdown of poly ribo­somes, indicating that lincomycin bound ribosomes cannot remain, attached to mRNA. It inhibits peptide bond synthesis in cell free systems, but probably has no effect on transpeptidation or termination in intact cells.
