Gap Closure


Gap Closure
- Closure of the gap converts the RFII form into the RFI form. The 5’ --> 3' exonuclease activity of DNA polymerase I removes the RNA primer remnant, and the DNA polymerase activity fills in the gap (simultaneously?). The 5' and 3' ends of the newly synthesized strand are finally joined by polynucleotide ligase.


The negative strand thus becomes a covalently closed circle. ØX174 and Ff parental DNA can give rise to two types of molecules. (i) Several copies of RF duplex are made by cellular polymerases to form a pool of progeny RF molecules (RF-->RF). (ii) Positive ssDNA strands are synthesized on a rolling circle.
