Infective Cycle - Eclipse

Infective Cycle - Eclipse -  During this stage there is irreversible loss of particle infectivity, and viral DNA is injected into the bacterial cell. A conformational change takes place in the phage particle. The phage binding site of ØX174 and 813 on the outer membrane of Salmonella typhimurium is a lipopolysaccharide, and appears to have the following structure (Jazwinski et. al, 1975)


The E. coli structure is analogous, but not identical, to this structure. It has been suggested that viral DNA is conveyed into the bacterial cell by the gene H protein which acts as a 'pilot protein'. This is supported by the fact that H protein has been found bound to the replicative form of on a after infection.


Reproduction of the ssDNA phage genome takes place in three stages:

(i) synthesis of a replicative form (RF),

(ii)replication of the parental RF to produce progeny RF DNA and

(iii) conversion of RF molecules into rolling circles.
