Okazaki Fragments
- The earlier studies of 14 replication based on petit phage particles led to the conclusion that replication was unidirectional from a fixed origin.

It was, however, subsequently established that replication was discontinuous (with initiation taking place at multiple origins) and bidirectional, at least for the first round.

Both strands are replicated discontinuously in the 5'-3' direction, leading to the formation of 'eye' structure. Synthesis of DNA takes place in short segments, the Okazaki fragments, which are then joined by a host ligase.
