Antisense RNA Strategy

Antisense RNA Strategy
The basic idea in antisense strategy is to block the expression of a particular gene product with the help of a transgenic construct containing the gene or part of the gene with the transcript in reverse orientation with respect to the promoter. This procedure results in the formation of a complementary RNA rather than the normal sense RNA.

This antisense RNA binds with its homologous sense RNA, preventing translation and/ or facilitating degradation or accumulation of a gene product by 90 to 99 per cent, thus creating a phenotypic mutant. There have been many reports of insertion of antisense construct of polygalacturonase enzyme into tomato plants. The resulting transgenic tomato plants had low levels of this enzyme and hence their fruits ripened slowly thereby increasing shelf life. Antisense RNA technology has also been used with success towards minimizing viral diseases.
