Procedures for AgrobacteriumMediated Transformations
Several procedures have been followed till to date: (a) through conventional gall formation on wounded plant stems or .leaves that are not amenable to monitoring with disarmed vectors, (b) Co-cultivation of Agrobacterium with plant protoplasts, a method that has been successful in plants where regeneration of plants from protoplasts has proved to be successful and (c) tissue transformation with ex-plants (leaf, cotyledon section or somatic embryos) inoculated with Agrobacterium, a procedure that has proved faster than protoplast regeneration.

When shoots are formed in regenerated plants, the plantlets are transferred to potted soil for acclimatization to natural surroundings.

The introduction of nif genes controlling nitrogen fixation in some microorganisms, into higher plants other than legumes is an experimental strategy envisaged by several research groups.
