Modulation
of
Complement
Systems
Several regulators modulate the complement cascade and prevent uncontrolled activation. Modulation is due to enzymatic instability of certain C intermediates and by specific inhibitors of certain components. Unstable enzymes are C4,2( C3 convertase)and C3b. Specific inhibitors are as follows :1. C1 esterase inhibitor, a normal 5erum constituent, inhibits the esterase activity of C1a by forming a firm reversible complex with it. Activation of C2 and C4 is thus inhibited.
2. C3b in activator present in serum cleaves C3b into two fragments, C3c and C3d. 3. Anaphylatoxin inactivator cleaves the C-terminal aringine from the C3a and C5a components and destroys their biological activities.
