Modulation of Complement Systems

Several regulators modulate the complement cascade and prevent uncontrolled activation. Modulation is due to enzymatic instability of certain C intermediates and by specific inhibitors of certain components. Unstable enzymes are C4,2( C3 convertase)and C3b. Specific inhibitors are as follows :­1. C1 esterase inhibitor, a normal 5erum constituent, inhibits the esterase activity of C1a by forming a firm reversible complex with it. Activation of C2 and C4 is thus inhibited.

2. C3b in activator present in serum cleaves C3b into two frag­ments, C3c and C3d. 3. Anaphylatoxin inactivator cleaves the C-terminal aringine from the C3a and C5a components and destroys their biological activities.
