IIA-IIB System - Sugar Specific Complexes - E. coli and S. typhimurium. Washed E. coli membranes were treated with urea and butanol, yielding a soluble fraction and a protein.

Both are required for phosphorylation of, fructose, glucose and mannose on addition of phospho-HPr.

Further purification of the soluble fraction gives three protein components. IIA Glc, IIA Man and IIA Fru. The pellet fraction on purification yields protein IIB in a lipid-free condition.

Reactivation of the protein requires both lipid (Phosphatidylglycerol of E. coli is most effective) and divalent cations
