Butt Joints Method
When breaks are introduced on the two DNA strands exactly opposite each other, there are no unpaired terminal nucleotides.

Such butt ends can be joined by bacteriophage T4 DNA ligase under appropriate conditions, provided that the 5' ends have terminal phosphate groups and that there is no blocking of the 3'OH groups, Insertions can take place in either possible orientation
