Phase Contrast Microscopy
Phase
Contrast
Microscopy allows
organisms
to
be
seen
alive
and
without
staining.
One
can
easily
see
the
internal
details
which
can
not
be
easily
visualised
with bright
field
microscopy.
Bacterial
granules,
fine
structure
of
protozoa
and
fungi
may
be
studied.
The phase
contrast
microscope contains
a
series
of
special
filters
and
diaphragms
that
split
the
light
beam
and
throw
the
rays
slightly
out
of
phase.
The
Separated
light
then
passes
around
microscopic
objects
as
wells
as
through
them,
and
small
differences
in
the
densities
of
the
objects
show
up
as
different
degrees
of
brightness
and
contrast.
