Electron Microscopic Method for Micormetry
In microbiology, specifically with the specimen containing bacteria and viruses, the optical methods for micrometry are now replaced by the technique involving the use of electron microscope. In this method, make a suspension of a suitable density of the microorganism to be measured and add to it a suspension of latex particles of known diameter 88 nm.
This mixture is used as the material for microscopy. Distortion of cells during microscopy is one of the major objections in the measurement. However, this can be avoided by careful fixation of the material. Measurements, since standard reference is also magnified to the similar extent.
