Electron Microscopy Freeze etching:
Electron micrographic investigations of unfixed living cells without artifact production in the result is the most striking feature of this technique. The procedure involves super cool­ing (-100°C) of a specimen soaked in 20% glycerine. Frozen block is then cleaved with the help of cool knife. Exposed sur­face is then etched, causing sublimation of the ice block. To give contrast to the etched surface, shadow-casting with metal is carried out. Carbon replicas prepared subsequently from this preparation are examined under microscope for the investi­gation.
