Electron Microscopy Preparation of section:
In principle, the preparation of sections for electron microscopy does not differ from that of light microscopy. However, to meet the specific requirements, modification is needed at every stage of process. Heavy suspension of bacteria or virus is required for this type of work which can be prepared by centrifuging them and suspending the pallet into the small amount of suitable medium. The process for the section preparation involves following steps. (1) Fixation, (2) embedding, (3) grid preparation and (4) ultramicrotomy.
