Electron Microscopy Shadow casting
This technique involves .a deposition of an electron dense material at an angle over a specimen. For this purpose various heavy metals like chromium, nickel, platinum, uranium, gold or their alloys are evaporated in the specially designed apparatus . Thus, the organism acquires a shadow on the uncoated side. This technique produces a topographical representation of the surface of a specimen. The method is more useful in examination of bacteria bearing flagella and pili and spirochetes.
