Method of Φrskov (19):
Molten nutrient agar is poured in the sterile petri plate to give depth of about 2-3 mm. A drop of culture suspension to be examined is placed in the center of the plate and spreaded by means of a bend glass spreader to give uniform inoculation. Small block of agar is cut out, using sterile scalpel and placed in the centre of a sterile slide in such a manner that the inoculated surface remains on the upper side. A strile cover glass is applied over the inoculated surface and is allowed to incubate till the desire period for observation.
