Modifications of hanging drop preparation
de Fonbrune (II) modified this method by introducing oil-bath to fill the air space as shown in Figure 3-2a. Oil bath fill-in preparations are successfully used to study the behaviour of the cells by various investigators (12-15). Another modification of hanging drop preparation is known as hanging block preparation, in which drop of cell suspension is sandwiched between the cover glass and thin agar block (Figure 3-2b).
This technique has serious drawback that the optical conditions are very poor due to the agar block and the air space. Elimination or air space by filling it with water or other medium has been suggested to form a hanging block with liquid in-fill to improve the optical conditions (Figure 3-2c).
