Modifications of hanging drop preparation
de  Fonbrune (II) modified this  method by introducing oil-bath to fill the air space as shown in Figure 3-2a. Oil bath fill-in preparations are successfully used to study the behaviour of the cells by various investigators (12-15). Another modification of hanging drop preparation is known as hanging block preparation, in which drop of cell suspension is sandwiched between the cover glass and thin agar block (Figure 3-2b).

This technique has serious drawback that the optical conditions are very poor due to the agar block and the air space. Elimi­nation or air space by filling it with water or other medium has been suggested to form a hanging block with liquid in-fill  to improve the optical conditions (Figure 3-2c).
