Observation of Microbes in Smeats:
Smear preparations are generally employed to observe bacteria and related organisms with respect to the size. Owing to the semitransparent nature of bacteria, though, impossible, but difficult to observe bacteria in an unstained condition. Stains are used in bacteriology with many objectives like; to increase the visibility of bacteria by making them contrast in colour with their surrounding, to see the shape and size of bacteria, to demonstrate the intracellular or extracellular structures of cell and to produce specific physical and chemical reactions. Staining reaction is very helpful in the classification and identification of bacteria.
