Paraffin Sections

Paraffin sections:
Blocks of material embedded in paraffin wax may be cut by the more conventional techniques. Routine bacteriological and mycological investigations of tissue is almost exclusively carried out with paraffin sections. The main objections to this method are due to the properties of paraffin itself and possible injury to tissue by high temperature during infiltration process

This can be successfully overcome by matching the hardness of paraffin, to the temperature at which material is going to be sectioned and the hardness of the tissue. The latter matching is also essential because warm paraffin shrinks as it cool and compresses the tissue in the block. Tissues harder than paraffin will only withstand this pressure. Difficulties may often come across due to the different properties of the microbes and the host tissue, but they can be successfully overcome by proper care during the fixation and dehydration procedures.
