Preparation and Fixation of Smears and Sections
Presence of microbes in the given specimen can be demonstrated either by direct microscopic observation or by culturing them on suitable media and the cultured specimen is latter examined microscopically. If the specimen is thick, it is necessary to take sections. In general, microscopic examination of bacteria is carried out by preparing smear from the culture suspension or liquid specimen, followed by staining the smear. Thus, preparation of smears and sections is an essential step in staining procedures. Preparation of material for electron microscopy is described at length in second chapter and accordingly we have devoted this chapter, only to the material preparation for the light microscopy.
