Preparation of Smears in Microscopy Method

Preparation of Smears in Microscopy Method:
Microscopic examination of bacteria has a great diagnostic value in medicine and is routinely performed by making suspension of specimen and staining the smear prepared from it. Smears' can be prepared on cover glass as well as glass slide. For making films on cover slip, Cornet's forceps are used to hold the cover slip into a horizontal position. Glass, slides are commonly used for smear preparation in bacteriology.

The sequence of steps needed to follow for the satisfactory result are as under. Take a clean grease free and dry slide. Make a circle of about 20 mm diameter with the help of glass marker. Deposit a drop of bacterial suspension to the reverse side of the marking in the centre of the circle. Spread the drop evenly as, thin film using sterile wire loop. A little experience will soon determine the amount and the density of the material required for this purpose. Allow the smear to dry in air at room temperature. Rapid drying can be ensured by warming the slide above the Bunsen flame. However, in some of the special staining techniques, preparation of smear requires extreme care.
