1. Wet mount or Wet Film Preparations
Beyond doubt the simplest form of microculture chamber is the wet mount preparation.They are prepared by enclosing the organisms in a layer of liquid between the slide and cover glass. There is ample room for the microorganisms to move while being observed and the results can be recorded perma­nently by the photographic attachment to the microscope. The major disadvantage of this preparation is that it dries out very rapidly. This can be minimised to some extent by sealing the edges of the cover glass by means of silicone grease to pre­vent the evaporation of liquid. Other disadvantage includes, the amount of medium that can be accommodated is very less.

This results into the condition of limited nutrient in a very short time. Oxygen starvation is the immediate effect upon obligate aerobes. To overcome these, the observation should be made at once. At the same time there are many advantages which have made this technique more popular and routine in microbiology. The preparations are easy to make and they are optically clean. One can use the highest resolving power to demonstrate these preparations because of their thinness. The deformation of light path is minimum as there need be only one layer of differing refractive index, if oil-immersion objective and condenser are used.
