Assay
In the excised root assay, acetylene reduction begins after 8-18 hours. Over night pre incubation of roots without acetylene at a low partial pressure of oxygen (pO2) permitted immediate detection of acetylene reduction.
The excised root assays result in higher estimates of nitrogen fixation compared with soil core assays, probably because nitrogen fixing bacteria increase in numbers during the pre incubation period.
