The Soil Dilution and Plate Count Method

This method has been generally used to estimate the number of microor­ganisms in the rhizosphere soil. From the data obtained, an es­timate of the total population of fungi, actinomycetes and bacteria per g of soil can be made.

The method of sampling the rhizosphere soil differs among workers and this could be one of the contributory factors for the observed variations in results obtained by different workers. Further, in the soil dilution and plate count method, spore forming fungi such as penicillia and aspergilli overwhelm the non-sporulating fungi especially those of basidiomycetes which thus often get overlooked.
