The Soil Plate
This method is an extension or adaptation of Warcup's soil plate method. The rhizosphere soil (0.05-0.01g) is directly plated on melted and cooled agar medium. Before the agar solidifies, the dishes are rotated to distribute the soil in the agar.
The advantage of this technique is that a differentiation of the soil microflora from various regions of the root system could be made. Secondly, mycelial as well as spore forming fungi have equal chances to come up on agar plates.
