When transmitted light instead of direct beam of light is used in a com­pound microscope, some structures in the specimen being examined ap­pear darker than others due to differential absorption of light by different parts of the specimen, thereby providing contrast not seen when viewed under direct light.

Both live specimens to understand motility of microorganisms as well as stained preparations can be viewed under a bright field microscope.

Wet mounts or 'hanging drop' preparations come in handy for observing live specimens. To prepare a hanging drop slide a depression in the centre of a glass slide is used and a drop of bacterial culture is placed in the depression.

Such depression slides are commercially avail­able. A coverslip is gently placed over the depression whose rim is covered with a smear of transparent petroleum jelly to seal the assembly. The depression side is turned over to face the microscope stage.

Simple wet mounts are also prepared by gently covering a drop of bacterial dilution with a cover slip and examining the preparation when it is still wet.
