Demonstration of Anthrax bacilli by simple staining:
In addition to simple staining, polychrome methylene blue solution (S21) is also used for the McFadyean’s reaction in which, anthrax bacilli are seen blue against purple granular surrounding. The slow oxidation of methylene blue forms a violet compound that gives this stain a polychrome property. Borax methylene blue reagent (S22) given by Masson demonstrates similar result as with polychrome methylene blue.
