(a)  Hiss Method (Modification of Anthony)
Anthony (15) modified the staining procedure described by Hiss (14). According to him, prepare a smear and do not fix it. Flood the slide with 1% crystal violet and allow it to react for 2 minutes. Wash off the stain with 20% CuSO₄, 5H₂O in water. Blot, dry and examine under oil-immersion objective. Cells appear dark purple while capsules are seen pale blue.
