Iron Haematoxylin Stain

(A) Iron haematoxylin stain
The schedule of staining is as under. Prepare wet smears as earlier described and fix with Schaudinn’s fixative (S40) for 5 minutes or longer, Wash the smear in 50% ethyl alcohol and apply Gram’s iodine (S24­) for 2 minutes to remove mercury salt. Rinse with ethyl alcohol till no more iodine comes away in the washing. Wash with distilled water and stain the smear with Weigert’s iron haematoxylin (S67) for 10-2 minutes.

Wash with distilled water and counterstain with Van Gieson’s stain (S68) for 15-30 seconds. Wash with water again, dry and observe. Sections are stained similarly upto this step then dehydrate rapidly with alcohol, clear in xylene and mount in Canada balsam. In this stain, amoebae and other protozoa are seen blue black in colour.
