Lendrums Phelaxine Tetrazine Method

(C) Lendrum's phelaxine-Tetrazine method
This method is useful in demonstration of acidophilic inclusion bodies. They are seen as red against yellow background. Nuclei take blue colour. Negri bodies are not stained. Staining schedule is as follows: Fix tissues in formol-saline (S61) or formol-corrosive (S62) solution and take paraffin sections as usual. Bring sections down to water.

Stain the sections in Mayer's haemalum (S64) for five minutes and then differentiate in water till they appear blue. Stain with 0.5% phloxine-B or 0.5% rose bengal in 0.5% CaCl2 for 30 minutes. Rinse in water and differentiate in saturated solution of tetrazine in 2-ethoxy-ethanol (cellosolve) by adding drop by drop. Rinse with 95% ethanol, dehydrate, clear and mount.
