Staining Method of Truant et al
This method (9) is advantageous when the number of organisms are less in sample. By this method a large area of sputum can be scanned in short time. The dye used for staining is fluorochrome dye. For the staining, prepare and fix the smear by usual method and stain fit with auramine staining solution (S28) for 15 minutes. Rinse the smear with water arid decolourizing the smear with decolourizing solution (S29) for 5 minutes. After washing with water apply 1 in 1000 potassium permanganate solution for 30 seconds to the smear. Wash,
dry and examine under 8 mn dry objective and a high power (20x) eyepiece. Tubercule bacilli will be seen as yellow luminous organisms in a dark field. Use of dark room will facilitate the observation. Oil-immersion objective is useful to check the morphology of acid-fast bacilli.
