In this in method heat treatment is not applied during the stain­ing, therefore, it is also known as cold method (13). Application of longer heat treatment during fixation makes stain permeable due to widening of pores of spore. The staining solution used here is also stronger and hence it stains spore.

Following below listed steps, one can perform the Staining. Fix the smear by passing through the flame 20 times and make the slide cool. Stain the smear with saturated aqueous mala­chite green (about 7.6%) at room temperature for 10 minutes.

Rinse the slide with water. Counterstain with 0.25% aqueous safranin solution for 2 minutes. Wash, blot dry and examine under oil-immersion objective. Spore appear green against red cell.
