Staining Method of Bouin

Staining Method of Bouin
In this method (28) cover slip impression smear is prepared. Fixation is used to fix the cells to cover glass. The sequence of steps is as follows. Remove agar slab of about 2 mm thickness on which organism is grown. Keep the cover slip in the centre of petri plate and place the agar slab onto it, facing growth side down. Add Bouin’s fixative (S12) into petri plate till the slab immerses in fixative. Keep it for 3 hours.

Carefully remove fixative first and then agar slab. Wash the cover slip by allowing water to drip into petri dish and over flow it for 2 hours. Treat it with 7% aqueous solution of tannic acid for 20-30 minutes. Wash the cover glass in water and stain with 0.02% aqueous crystal violet for 1 minute. Mount in water and seal with nail varnish. Bacterial cytoplasm will appear pale purple, while cell as dark, often brownish black. Cross walls are readily visible.
