Staining Method of Burdon
The schedule of staining described by Burdon (10) is as shown below. Prepare a smear and fix it by heat. Flood the slide with sudan black-B staining solution (S46) and allow it to react for about 10-15 minutes. Drain off excess of stain and dry it. Do not wash with water. Rinse the smear with xylene and blot dry. Do not wash with water. Counterstain with 0.5% safranin or with dilute carbol fuchsin (S19) for 5-10 seconds. Rinse with water, blot dry and examine under oil-immersion objective. Lipid granules appear deep blue against light pink cytoplasm
