Staining Method of Castaneda Modification of Bedson Procedure

Staining Method of Castaneda modification of Bedson's procedure
This procedure is useful for rickettsiae as well as for ele­mentary bodies of psittacosis. They are stained blue, while cell nuclei and cytoplasm appear red. For the staining, prepare the film from infected tissue and dry in air. Fix the film in 1N HCI for 2 minutes and wash thoroughly to remove acid. Stain the film with azure-II solution (S65) for 20 minutes.  Wash with distilled water and counterstain with 0.25% safranin not more than 6-8 seconds. Wash in running water, blot
and dry.
