Staining Method of Dobell
The method described by Dobell (21) is useful for staining intestinal amoebae and other protozoa. In this method fix the smear in Schaudinn’s fluid (S40) for 5 minutes. Wash with distilled water. Apply 2% aqueous ammonium molybdate for 10 minutes as mordant. Wash again with distilled water. Stain the film for 10 minutes with unriped, freshly prepared haematoxylin solution (S67). Wash with distilled water and transfer the film into tap water for about 30 minutes or longer until the film appear blue in colour. Mount in water. Sections are treated similarly, but dehydrate with alcohol, clear with xylene and mount in Canada balsam. Amoebae and other protozoa appear blue in colour.
