Staining Method of Doner with Synders Modification

Staining Method of Dorner with Snyder's modification
The method given by Snyder (12) is as follows: Prepare a smear and dry it in air. Cover the smear with blotting paper and saturate it with carbol fuchsin (SI9). Heat the slide from below till steam rises for 5-10 minutes. Keep the blotting paper moist by adding stain dropwise during the process. To obtain neat preparation, decolourize immediately with 95% ethyl alcohol, but omit this step if organisms do not hold colour well

Rinse the slide. Place a drop of saturated aqueous nigrosin at one end of slide and spread it evenly with the help of another slide. Dry the slide quickly using gentle heat and observe under oil-immersion objective. Spores will appear red while background of cell black, leaving the cell colourless.
