Staining Method of Dyar

Staining Method of Dyar
Cetylpyridinium chloride dissociates in water to form positively charged cetylpyridinium and negatively charged chloride ions. As a result of absorption of cetylpyridinium cations on bacterial cell wall, it becomes positively charged and on subsequent treatment with acidic dye like congo red, cell wall is stained, while cytoplasm is stained with basic dye like methylene blue.
The method describe by Dyar (29) is as follows:

Prepare the smear, air dry and fix it by heat. Place 3 drops of 0.34% aqueous solution of cetylpyridinium chloride on the smear. Add I drop of 5% congo red. Mix and wash with water.

Cover the smear with 0.5% methylene blue solution and allow, it to react for 10 seconds. Wash, dry and examine under oil-immersion objective. Cell wall appears red in colour while cytoplasm blue in colour.
