Staining Method of Field
The method described by Field (23) can be used as an alternative to the Giemsa's method for thick blood films. Prepare a thick blood film as earlier described (page 57) and dry in oven at 40°C. Pass the film rapidly two to three times, having each passage of 2-3 seconds duration through the Bunsen flame. Cool the slide and immerse in Field's stain-A (S72) for not longer than 1-2 seconds. Rinse the slide gently with distilled water until colour ceases to flow from the film and the glass of slide is free from stain. Immerse the slide in Field's
staining solution B (S.73) for 1-2 seconds only. Rinse gently in distilled water for 2-3 seconds. Place the slide vertically against a rack to drain and dry.

To get better differentiation of 2-3 weeks old films and unduly thick films, they may he immersed in buffer solution (S70) prior to staining until haemoglobin begins to diffuse out.

For the differential white blood cell count, the method described as follows is useful. Prepare thin blood films and fix in methanol for 5 minutes. Stain for 40 minutes in the staining solution prepared by mixing Giemsa stain stock solu­tion (S71), 2 ml; buffer solution (S70), 4 ml and distilled water, 94 ml. Wash gently in distilled water till smear becomes pink. Blot gently, dry and examine.
