Staining Method of Gray
This technique the modification of Muir's method and was proposed by Gray in 1926 (25). This method gives the cleanest background of all the flagella staining, where flagella appear light red and cells red in colour. Staining can be done by preparing a smear as described earlier. Pour Gray's staining solution (S30) on the smear and allow it to react for 10 minutes at 25°0. Wash gently with distilled water. In option, counter staining with carbol fuchsin for 5-10 minutes may be employed. Wash with water, air dry and examine under oil-immersion objective.
