Staining Method of Guststein

Staining Method of Gutstein
This method (14) is quite useful for staining the elementary bodies of variola-vaccinia group of viruses in smears made from scrappings of skin lesions. The schedule of staining is as under. Prepare thin film and remove protein by rinsing in saline and then with distilled water.

Fix the film in methyl alcohol for 30 minutes. Support the slide, film facing down wards on two capillary tubes placed in petri dish. Add Gut­stein's stain (S64) in the petri dish, cover the lid and allow the stain to act for 20-30 minutes at 37°C. Remove the slide, rinse with distilled water and air dry.
