Staining Method of Levaditi

Staining Method of Levaditi
This method is used to detect spirochetes in tissue. This is the modified method (5) of Becker's in which pyridine is used. This method is rapid and gives better result. The sequence of steps is shown below. Fix a small piece (1 mm thick) of tissue by keeping it in 10% formalin for 24 hours. Wash the tissue in water for 1 hour and then place it in 96-98% alcohol for 24 hours for dehydration. Place the tissue in silver nitrate pyridine solution (S56) for 2 hours at room temperature.

Transfer the container at 500C and keep at this temperature for 4-6 hours.  Wash the tissue in 10% pyridine solution and rapidly transfer it to reducing fluid (S57) and keep it for 48 hours at room temperature in dark.  Wash with distilled water, dehydrate in increasing strength of alcohol.  Take sections and after removing paraffin with xylol, mount sections immediately on Canada balsam.
