Staining Method of Macchiavells for staining rickettsiae
This method is advantageous for staining rickettsiae in films from tissues. The staining schedule is as follows. Prepare the film from tissue and air dry.  Warm the slide gently and stain for 4 minutes with 0.25% aqueous solution of basic fuchsin (pH 7.3) Filter the stain before use. Wash rapidly, first with 0.15% citric acid and then with water. Counter­stain with 1% aqueous methylene blue for 5 seconds. Blot dry and examine.
