Staining Method of Moller
The following steps are recommended for this method (17). Prepare the smear and fix it by Moller's fixative (S32,) for 15 seconds. Drain the fixative and blot dry. Cover the smear with crystal violet solution (S33) for 1-3 minutes. Wash the smear with saturated CuSO4 5H2O solution for 1 0 seconds . Blot dry and examine under oil-immersion objective. Capsules appear light purple violet, while bacterial cells as dark violet in colour.
