Staining Method of Nicholau

Staining Method of Nicholau

This method has advantage in detection of viruses in thick film containing high protein. Staining solution is also very stable and can be kept for indefinite period. Fix the smear with gentle heat or methanol. Cover the smear with Nicholau's staining solution (S66) and heat the slide from below till steam rises for 5 minutes. Ensure that the stain does not boil.  Rinse with water, blot dry and examine.
