Staining Method of Rue

The technique, described by Rue (4) gives better results than the method of Fontana.  In this technique, the apparent thickness of organism is increased by application of basic dye which forms precipitates in the presence of mordant like tannic acid, sodium bicarbonate or potassium permanganate and settles onto the cell surface.

Schedule of staining is as follows: Prepare a smear and air dry.  Fix the smear with 1% formalin for 1 minute and dry.  Place a drop of 5% NaHCO3 and then add 10 drops of basic fuchsin (S55).  Mix well and stain for 3-5 minutes at room temperature (observe visible precipitates).  Wash, dry and examine under oil-immersion objective.  Spirochetes appear red in colour.
