Staining Method of Webb

Prepare a smear and fix it by heat. Flood the smear with 5% aqueous solution of tannic acid for 30 minutes. Rinse the slide with water. Do not dry. Stain the smear with 0.5% aqueous solution of crystal violet chloride for 2-3 minutes.

Wash off the stain with water and treat the smear with 0.5% aqueous solution of congo red for 2-3 minutes. Wash, dry and examine under oil-immersion objective. Cell wall including newly developed cross walls and completed transverse septa will be stained blue.
