Modified method of Bailey

The method described here is, the method of Bailey modified by Fisher and Conn (23). Prepare the smear as earlier described. Filter solution-A (S36) onto the smear and allow it to react for about 3-5 minutes. Pour off solution-A and, without washing add freshly filtered solution-B (S37). Allow it to react for 7 minutes. Wash carefully with distilled water.

Before slide dries, cover the smear with Ziehls fuchsin (S19) and heat it from below till steam rises for I minute. Rinse the slide with water. Air dry and examine under oil-immersion objective. Flagella and cell will appear red, background precipitates are usually finer and less conspicuous.
