Negative Stains for capsules
Capsules are also easily demonstrated by negative staining technique. Due to the non-ionic nature of capsule, it has very little affinity for dye. Therefore, when dye solution is applied, it stains the cell and on application of nigrosin background is stained leaving capsule colourless. Thus, cells are positively stained and capsules are negatively stained.
