Staining of colony:
The most useful and routine stain is that of Dienes (11) modified later by Madoff in 1959 (12). The schedule of staining technique for direct observation is as under. Flood the plate or portion of plate containing suspected colonies of mycoplasma with 1:9 diluted Dienes' staining solution (S58) in distilled water. Examine the plate under microscope using low power objective.
The colony will appear granular and extends through several focal planes, giving rise to the appearance as collection of several cells of royal blue to greenish blue colour
