Standard Plate Count (SPC)

Standard Plate Count SPC

Water sample is diluted in sterile buffer. Measured amounts are pipetted into Petri dishes. Agar medium is added and plates incubated. Colony counts are made and multiplied by the reciprocal dilution factor to have total bacteria per ml water. This method is similar to that used for milk.
